[Abstract]ObjectiveToinvestigate theefectof enriched environment(EE)onthe motorand cognitivefunctions of mice with middlecerebralarteryocclusion/reperfusion(MCAO/R)and itsunderlying mechanism.MethodsAmouse model of MCAO/R wasconstructed,andthen1daylater,the micewerefedinEEorstandardenvironmentfor14days.The modified Neurological Severity Score(mNSS)and therotarod test wereused to evaluate the motorfunctionof mice;theY-mazetest wa used toevaluatethecognitivefunctionofmice;Westernblotingwasusedtomeasuretheexpresionlevelofvesicularglutamatetransporter1(VGLUT1)in thecortex,striatum,and hippocampusof theipsilateral side.ResultsEE significantlyreduced the mNSS score of MCAO/R mice ( ⋅F=103.900,q=9.551,P<0.001) and increased fall latency in the rotarod test,the percentage of time spent in the novel arm,and the percentage of entries into the novel arm in MCAO/R mice Δ(F=10.110-31.910,q=4.271- (204号 7.769,P<0.05) ,and it also reduced the expresion level of VGLUT1 in thecortex,striatum,and hippocampus of the ipsilateral hemisphere of MCAO/R mice (F=8.141-14.600,q=5.318-7.240,P<0.05) .ConclusionEE can significantly improve the motor and cognitive disorders in mice with MCAO/R,possibly by downregulating the expression of VGLUT1.
