[Abstract]ObjetiveToinvestigatetheeffctsof intranasal administrationof ferricammonium citrate(FAC)onthe expresion of glucocerebrosidase(GCase)andlysosomalintegral membrane protein 2(LIMP-2)in mouseolfactorybulbs.Methods Forty-eight male C57BL/6 mice were randomly divided into 6 groups with 8 mice in each group. Three groups were given FAC bilateral alternate nasal drops (200mg/kg )once every other day for1,3,or 6 weeks,respectively.The 3 control groups were given the sameamountofnormalsalineoverthesametimeperiods.Proteinlevelsof lghtchainferrtin,GCase,andLIMP-2inolfactory bulbs weredetected by Western blotting.ResultsCompared withthecontrol groups at1,3,and6 weeks,the expressionof lightchainferitinintheolfactorybulbsofmiceintheexperimental groupsatthecorrespondingtimepointswassigniicantlyincreased Δ:t=6.289-27.270,P<0.001) . There were no significant differences in GCase protein expression between nasal administration of FAC and nasal administration of saline at 1,3,and 6 weeks ( P>0.05; .Compared with nasal administration of normal salinefor1week,theexpressionofLIMP-2inofactorybulbsofmicegivennasaladministrationofFACfor1wkwassignificant lyincreased (t=4.483,P<0.01) ,but there were no significant differences in the expression of LIMP-2 at 3 and 6 weeks ( P> 0.05).ConclusionTherewassignificantirondepositionintheolfactorybulbsafternasaladministrationofFAC,butitwas insufficient to induce significant dysfunction of GCase or LIMP-2.
