Effects of neuropeptide FFon proliferation,invasion,and migration of invasive ductal carcinoma cellsCU Yujie,WANG Ting ting,WANGYe,ZHANGChunling,PANG Zongxin(School of Clinical Medicine,Qingdao University,Qingdao 266073, China)

[Abstract]ObjetiveToinvestigate theeffectsof neuropeptideFF(NPFF)ontheproliferation,invasion,and migrationof invasiveductalcarcinoma(IDC)celsand theasociated mechanisms.MethodsWe measured thelevelof NPFF in IDCtissue by enzyme-linkedimmunosorbent assay;determined theexpresionofNPFFandNPFFreceptor2(NPFFR2)inIDCtissuebyimmunohistochemistry;measured NPFF levelsin IDCcelIines(MCF10A,MCF10AT,MCF10DCIS,and MCF10CA1d)by Western blot;assessedtheefects ofNPFFontheproliferation,migration,andinvasionof MCFoAcelsby CellCounting Kit-8assay, scratchassay,andTranswellassay;andmeasuredtheexpressionof proteinsrelated totheextracelllarsignal-regulatedkinase (ERK)1/2 signaling pathwaybyWesternblot.ResultsCompared withthoseinadjacenttissue,theexpresionlevelsof NPFF and NPFFR2 proteins in IDC tissue were significantly increased ( ,both P<0.001 ). The expression of NPFF protein in IDC tissue was significantly higher than that in ductal carcinoma in situ tissue ⟨t=5.34,P<0.001⟩ . MCF10CA1d cells had significantly higher NPFF and NPFFR2 protein levels than MCF1oA cells ( F=9.92,6.64 ,both P<0.05 ).Compared with NPFF at 0μmol/L ,NPFF at 20 and 40μmol/L significantly increased cell viability and migration,and NPFFat 80μmol/L significantly increased cell invasion and p-ERK1/2 protein expression Ω(t=4.72,5.84,F=3.56 ,all P<0.05 );and cells treated with NPFF at (204 80μmol/L plus BIBP3226 at 80μmol/L exhibited significantly decreased invasion ability compared with the 80μmol/L NPFF group ⋅P<0.05 . NPFFR2 knockout significantly reduced the activation of ERK1/2 induced by NPFF at 80μmol/L (t=6.43,P< 0.05). ConclusionNPFF is highly expressed in IDC tissue,which promotes the proliferation,migration,and invasion of IDC

cells by activating the ERK1/2 signaling pathway.
