维替泊芬对脑缺血损伤早期脑组织和星形胶质细胞的影响

[Abstract] Objective Toinvestigatetheefectofverteporfin(VP)onbraintisueandastrocytesintheearlystageofcerebralischemicinjury. Methods Atotalof36Sprague-Dawleyratswererandomlydividedintosham-operationgroup (Sham group),midlecerebralarteryoclusionmodelgroup (MCAOgroup),andVPtreatmentgroup (MCAO+VPgroup),with12 ratsineachgroup.TheratsintheMCAOgroupandtheMCAO + VPgroupwereusedtoestablishamodelofMCAO,andafter modeling,theratsintheMCAO + VPgroupweregivenintracerebroventricularinjectionofVP (0.1mg/kg) ),whilethoseinthe Shamgroupweregivenshamoperationalone.The2,3,5-triphenyltetrazoliumchloride(TTC)stainingmethodwasusedtoobserve cerebralinfarctionineachgroup,andimmunofluorescenceasaywasusedtoobservethechangesinglialfibrilaryacidicprotein (GFAP)andYes-asociatedprotein(YAP)inastrocytes. Results TTCstainingshowedthatonday3ofcerebralischemicinjury,theMCAOgroupandtheMCAO+VPgrouphadasignificantlyhigherinfarctvolumethantheShamgroup,andcompared withtheMCAOgroup,theMCAO+VPgrouphadasignificantreductionininfarctvolume ΔF=201.00,P<0.05) ).Theresultsof immunofluorescenceasayshowedthattheMCAOgrouphadasignificantlyhighernumberofGFAP-positivecelsthantheSham group,andcomparedwiththeMCAOgroup,theMCAO + VPgrouphadasignificantreductioninthenumberofGFAP-positive cels ′F=8.51,P<0.05) ),aswelasasignificantreductioninthenumberofYAP-positivecelsamongtheGFAP-positivecels ⟨F=78.41,P<0.01) ). Conclusion VPexertsaneuroprotectiveefectintheearlystageofcerebralischemia.
