[Abstract] Objective To investigate the efects and mechanism of Zhongye Zhixie Mixture （ZYZXM）on mice with spleendeficiencydiarhabasdontheToll-likereceptor4/myeloiddiffrentiationprimaryresponseprotein8/uclearfactorkappaB （TLR4/MyD88/NF-KB）pathwayand gutmicrobiotaMethodsGavage administrationofrhubarbdecoction wasperformedtoestablish a spleen-eficiencydiarhea model SixtymaleKMmice wererandomlydividedintosix groups，includingablank group，a model group，a positive druggroup，andhigh-，medium-，andlow-dosegroupsof ZYZXM，with10miceineach group.Aftersuccessful modelestablishment，eachdrugadministration groupreceivedcoresponding medicationvia gavagefor7days.Thebodyweight， diarrheaconditionandsplenindexofthemicewerecomparedbeforeandaftertreatmentHematoxylin-eosin（HE）stainingwas usedtoobserve the morphologicalchanges inthecolontissuesof miceincach group.16SrDNA high-throughputsequencing was employed tocheckalterations inthegutmicrobiotastructureofmiceineachgroup.Enzyme-linkedimmunosorbentassy（ELISA） was utilized to measure the levels of interleukin-6 （IL-6） and tumor necrosis factor- α （TNF- ∇⋅αα ） in the serum of mice in each group. Westernblotanalysiswasperformed toexaminetheproteinexpressonlevelsofTLR4andMyD88 inthecolontissesof micein eachgroup.Immunofluorescence wasusedtocheckthenuclear translocationof nuclearfactor-kappaB（NF-kB）p-p65proteinin thecolontisues of mice ineach group.Results Compared withtheblank group，themice inthemodel groupexhibiteddecreases in body weight and spleen index （ P κκκ1 号 p-p65 in colon tissues （ Pe 0.05）.
