Genome-wide identification and expression analysis of the asparagine synthetase gene family in wheat

Abstract:Basedonthe proteinsequences oftheasparagine synthetase(ASN)genefamilyfrom Arabidopsis and maize,agenomewideidentificationoftheASNgenefamilyinwheat(TriticumaestiumL.)wasconductedusingbioinformaticsmethods,andacoprehensiveanalysisofthesicochemicalpropertis,onsereddomains,hromosomalocations,ndylgeneticelatisisofits memberswasperfored.Meanhile,theexpresionpatersofASNgenesindiferentwheatorgans,atvariousdevelopmentalstages, andunderbioticandabioticstreses weresystematicallyanalyzedusing transeriptomedata.Theresultsshowedthatall14identified wheatASNfamilymemberswerehydrophilicproteinsanddistributedon11chromosomes.Subcelularlocalizationpredictiondicated thatwheatASNproteinswereprimarilylocatedinthecytoplasm,folowedbythecytoskeletonandmitochondria.Phylogeneticanalysis revealed thatthe wheatASgenescouldbedividedintothre subfamilies,andtheconservedmotifs weresimilaroridenticalamong memberswithinthesamesubfamily.Cis-actingelemenanalysisidentifiedeighttypesofregulatoryelementsinthepromoterregions, mainlyncludingabsisicidsposiveess,erellsposives,oughtiducibilitlightsposiveness,nda olismelements,suggesting tattheheatASNfamilygeneswereinvolednotonlyinresponsestovarousstressesbutalsoinmultiple hormoneregulations.AnalysisofabioticstressresponsesshowedthatunderNaCltreatment,theexpresionofallgenesexcept Ta ! ASN11 was down-regulated.UnderPEG-siulateddroughtstress,theexpreionofTaASN1,aASN7,andTaASN9 wasdowegulated,while thatofTaASN8,TaASN10,andTASN1wasup-regulated.AfterABAtreatment,,theexpresionofTaASN9wadown regulated,whereas theother geneswereup-regulated.The14 wheatASNgenefamilymembers wereevolutionarilyonservedand widely involved in the regulation of biotic and abiotic stress responses.
