Abstract:ObjectiveToanalyzethediferentialexpressionandbiologicalfunctionsofcircRNAsintheanteriorlenscapsulesof highmyopicpatients withcataractandtheirpathogenicrolesinthedevelopmentofthiscondition.MethodsAnteriorlens capsulespecimens werecolected intraoperatively from36patients withage-relatedcataract (ARC)and36highmyopic patients withcataract.Among these,18specimens fromeachgroupwereselected for wholetranscriptomesequencingand biologicalanalysis,and theremaining 36 specimens wereused for validationofcircPDGFRA,circFOXJ3,hsa_circ0004767, hsa_circ_0007528,ciCRIM1,circMAN1A2,circSLC5A3,andcircPTK2expressionsusingRT-qPCR.hsa_circ0007528was selectedforcellexperimentstoexamineitsectsonproliferation,migration,andapoptosisoflnsepithelialcs (s). ResultsA total of16192 circRNAs were detected inthe specimens from both groups,among which 62 circRNAs were diferentiallyexpressed(29upregulatedand33downregulated).GOandKEGGanalysesrevealedthatthediferentialy expresedciRAswereprimarilylocalizedinhecoplasm,uleoplasmandedoplasmicticulum,andweeinvoled insignalingpathwaysassociatedwithGapjunctionandtheP3K-Akt,NF-B,Jak-STAT,HF-1andMAPKsigaling pathways.TheceRNAnetwork predictedmultipletarget genes.RT-qPCRvalidationresultswereconsistentwiththe sequencingdata.IntheLECs,upregulationofhsa_cic0o7528significantlyinhibitedcellproliferationandmigratioand obviouslypromotedcellapoptosis.ConclusionTheexpresionprofileofcirRNAsintheanteriorlenscapsuleofhighmyopic patientswithcataract differs from thatofARCpatients.Upregulationof hsa_circ_0o07528 inhibitsLECproliferationand migration and promotes cell apoptosis.
