Abstract： ObjectiveToinvestigatetheexpressionlevelsof marginalzoneBandB1-cellspecificprotein （MZB1）inpan-cancer anditsassociation with patient prognosisand tumor microenvironment （TME）.Methods MZB1 expression data， clinicopathological parameters，and survival data from 33 cancer types were extracted from the UCSC database foranalyzing thecorelationsofMZB1 withclinical stage，patient prognosis，immunomodulatorygenes，immunecheckpoint genes，tumor stemnessimecelliilraomouaoalen）doselitsbility.Zio in pan-cancer were asessd usingcBioPortalonlinedatabase，andthevalueofMZB1forcancerdiagnosis was evaluatedusing ROCcurveanalysis.MZB1expressionlevels inmyeloidleukemiaandrenalcarcinomacelsweredetected usingRT-qPCRand WesternblottingandtheefectofMZB1knockdownoncellproliferationwasexaminedusingEdUassayResultsMZB1was significantlyoeexpressedin2certyesludingideynalarcellrcioa（KC）astinasieoa andacutemelidka.Ieiossoiadithgealageallialndio free survival inmultiplecancers.Inmost tumors，，MZB1 expression wascorelated significantlywithimmunomodulatory genes，immunecheckpoint genes，tumorstemness，immunecellinfiltration，TMB，andmicrosateliteinstability.Gene amplificationwas the predominant mutation typeof MZB1in pan-cancer，and MZB1 showed high diagnostic value forskin cutaneous melanoaKICandadandnecksqamouscellarcinoa.MZB1wasighlyexpressdindieenteloi leukemiacellinesandrenalcarcinomacellines，andMZB1knockdownsignificantlysuppressed theproliferationofH60 and769-Pcell.osionZBisiglexpesedinetyfmoranditsabatexpriofctste and prognosisof many tumors，suggesting itspotentialasanoveltumor biomarker andimmunomodulatorytarget.
