Study on Tissue Culture Technology of Syringa vulgaris

AbstractTaken stem segment of Syringa vulgaris as experiment material， the optimal culture conditions based on disinfection，primary culture，secondary proliferation culture，rooting culture and domestication transplantation by disinfectant，disinfection time，basic medium，hormone ratio and transplanting matrix were carried out.The results show that，the best disinfectionmethodwas 75% alcohol 15s+1%HgCl24min. the survival ratewasashigh as 84% ;with stem segment as the explant material.The best base medium for the primary culture was MS medium，the optimal medium for the primary culture was MS+2，4-D 0.1 mg/L+6-BAl.0mg/L . Suboptimal medium for proliferation was MS+6 -BA 0.5mg/L+NAA 0.2mg/L， the proliferation coefficient was 5.65，The optimal rooting medium was 1/2MS+IBA2.0mg/L+NAA0.2mg/L， the rooting rate was 84.9% .Root seedlingswere domesticated and transplanted to vermiculite，the survival rate was 82.2% ：
