[Abstract]ObjectiveTo prepare a wound dressing with rapid hemostasis and sustained antibacterial activity.Methods Cuprous oxide nanoparticles were mixedwith chitosan-gelatin(CG)solutionto preparecuprous oxide-chitosan-gelatin(CuCG) freze-dried spongeusingthefreeze-drying method.Aseriesofexperimentswereperformed toassessthephysicochemicalpropertiesofCuCGsponge,suchascoagulationtime,scannngelectronmicroscopyinfrared spectroscopy,swelingbehavior,waterre tentioncapacitycompresionstrength,ndantibacteralactivityRatmodelsofliverijuryndinfectedfullthickneskidefect wound wereusedtoobservethehemostaticandantibacterial efectsofCuCGspongeinuiuo,whichwascomparedwithchitosan (CS),and CG sponges.ResultsCompared with CSand CG sponges,CuCG sponge dressing had significantly higher porosity 1 ∵F=6.64,P<0.05) ,swelling rate ( F=6.07,P<0.05) ,and compression strength ( F=8.51,P<0.05) ,with good antibacterial activities against Sta phylococcus aureus ( F=475.80,P<0.001) and Escherichia coli( F=73.36,P<0.001) .In uivo experiments showed thatcompared withthemedical gauzecontrol groupandtheCG sponge group,theCuCG grouphad significantlybetter time to hemostasis ( F=4.68,P<0.05 )andbleeding volume (F=16.16,P<0.01) for liver injury in rats,and on days 7 and 14, there was a significant diference in would healing rate between the CuCG group and the other two groups ( ′F=5.13,4.72,P< 0.05).Conclusion The novel CuCG dressing prepared in this study has good hemostatic and antibacterial effects,and therefore, it has a potential application value in war,natural disaster,and other situations.
