Preliminary study on virus-free seedling culture and container potato induction technology of Mengwushu No. 6

Abstract: Using virus-freeseedlingsof the new drought-resistant and high-starch processing potato variety Mengwushu No.6asexperimental material,this study investigatedtheeffectsofdiferentcultureveselsonthegrowthof variety virus-free seedlings,screened theeffectiveconcentrationof Yipeilong bacteriostaticagent forbacterial contamination control,andexploredthesuitableinductionmethodforcontainerpotatoofthisvariety.Theresultsshowedthat the virus-free seedlings in the fullautomatic plastic square box were more robust, with improved photosynthetic efficiency and stronger growth potential.This treatmentcould increase the multiplicationcoeficientofpotatovirus-free sedlings, which was superiortothe glassbotle culture.The most suitableconcentrationof Yipeilong forpreventing bacterial contamination in Mengwushu No.6was 0.20% .Therapid induction culture method forin vitro potato tubersisas follows:Solid medium(seedling- strengthening stage: MS+30g⋅L-1 sucrose+6 g⋅L-1 carrageenan) + liquid medium(container potato induction stage: MS+80g ·Lsucrose +500mg⋅L-1 chlormequat chloride+5 mg L-1 6- benzylaminopurine).The resultsofthis studynotonlylayatechnicalfoundationfortheindustrializationdevelopmentofMengwushuNo.6,butalso provide a reference for research on similar potato variety.
