Abstract:Toclarifythe effects ofdiferent types ofrootstocks on thequalityof watermeloncultivated in greenhouse and screensuitablerootstocks forwatermeloncultivationinNingxiafacilities,Ningmei2(NM2)watermelonwasusedas scion,and Ningzhen2(pumpkin,NZ2),inggen(gourd,QG),Ningzhen3(wildwatermelon,NZ)arootstocks,toinvestigate post-grafting changes in watermelon quality.Bycombining transcriptomic and metabolomic analyses,the influenceof thethree rootstocktypes onwatermelonqualitywascompared.Theresults showed that diferentrootstock types significantlyectedgeneexpresion,pysioogicalandbiochemicalproperties,andfruitqualityofwatermlocultivatedingreenhouse.Among them,NZ2 grafting increased thecentral soluble solidscontentby0.2 percentage points, and the vitamin C content by 2.7% . Italso significantly influenced protein processing pathways,potentially enhancing quality by bolstering natural defense mechanisms.QG grafting hadthe most significant influence on gene expression and metabolitechangesbutresultedinslightlyinferiorfruitquality.NZ3grafted watermelonexhibitedthehighestcrudefiber content, 65.79% higher than that of NM2.The application of transcriptomic and metabolomic techniques to study the changesoffruitqualityaftergrafting providesatheoreticalbasis forscientific screning ofrootstocksandimproving the quality of grafted watermelon.Through comprehensive analysis,the pumpkin rootstock NZ2 was considered asa suitable rootstock variety.
