Comparison of ploidy identification methods for regenerated plants from pepperantherculture

Abstract:Inorder toexploreanacurate,eficient,andapplicable ploidyidentification method forregenerated plants from pepperantherculture,inthisstudy,6Oregenerated plantlines from1O genotypesofpepperantherculture wereused as the test materials.The ploidy identification was conducted by adopting the plant fertilitydetection method and DNA flowcytometry,combined withthechromosomecounting methodofroot tipcellsquash.Theadvantagesanddisadvantagesof diferent methods werecompared.Theresults indicated thatthe accuracyrateof ploidy identification byDNA flow cytometry was 96.67% ,this method was rapid and convenient,but it had highcost and was prone to peak bias.The pollen vitality detection method had an accuracy rate of 100% in determining the odd-even ploidy of regenerated plants' chromosomes,but itcouldnotdistinguishthe specific ploidyofregeneratedplants.Thecombinationofpolenvitalitydetection method and DNA flowcytometry detection method could significantlyreduce the identificationcost,improve theaccuracyrateandeficiency.Thatis,forthelines thathavealreadyflowered,thepollenvitalitydetectionmethodcouldbeused to determine theodd-even ploidy of chromosomes,which was convenient for early chromosome doubling treatment; for thelines thatfloweredlater,theDNAflowcytometrydetectionmethodshouldbeused inthesedlingstage.Ifthere were difficult-to-determine situations suchaspeakbias,thepollnvitalitycouldbecombined todetermine the ploidyofregenerated plants.
