Establishing of a wet-cold tolerant identification method for cucumber during seedling stage by wet-cold injury index

Abstract： To establish a wet-cold tolerant identification method during seedling stage for cucumber by morphology，the seedlings intwodevelopedtrue leavesof8testcultivarsand5validationcultivars withdiferent wet-coldtolerance were treatedunder 9∘C/5∘C（day/night）+ relative humidity 95% growth chambers（taking 25∘C/18∘C +relative humidity 85% asthecontrol）.After7daysof treatment，thedegreeof wet-cold injurywascountedforeach plant，the wet-coldinjuryindexes were calculatedand the growth parameter were measured.The results showed thatthe diferent cultivars presented diferentlevelofleafyellowing，waterlosing wiltingordeath underwet-coldstressandthe wet-coldinjuryindexes could indicate the wet-cold toleranceofthecultivaraccurately.Theresultsofvalidation testwereconsistent withthoseofmethodestablishment test.Theclusteringofwet-cold toleranceofvarious varietiesbasedontherelativevaluesofplantheight， leaf lengthand leaf width etc.under wet-cold stress and controlcondition wereconsistent with theclasificationof wet-coldinjuryindex.Therefore，itisthoughtthatthewet-coldinjuryindexatseedlingstagecanbeusedtoidentifythe wet-coldtoleranceofcucumber cultivars.The wet-cold injury index greater thanObutlessthanorequal to0.250 ndicatesstrong toleranttype，greaterthanO.250butlessthanorequalto0.550indicatesmoderate toleranttype，greaterthan 0.550 butless thanorequalto0.750indicates weak tolerant type，andgreaterthan0.750butlessthanorequalto1.00 indicates sensitive type.
