Transcriptome sequencing and bioinformatics analysis of melon seedlings under waterlogging stress

Abstract:Waterlogging isoneof themost widespreadand frequent natural disasters worldwide.Asacucurbit crop,melon exhibits relatively poor waterlogging tolerance.When melon cultivation areas experience prolongedrainfallor flooding,severeyieldloses orevencompletecropfailureoftenoccur.Inthisstudyawaterlogging tolerant melonhighgeneration M13-06-3,wasused as experimental material. Transcriptome sequecing and quantitative real-time PCR(qRT-PCR) were employed to analyze diferentially expressed genes(DEGs)inroot tissues.The results revealed that 9037 DEGs were screned in M13-06-3 under waterlogging stress including 4529 upregulated and 4508 downregulated genes. qRT-PCR validationconfirmed that the expression patternsof five genes,MELO3Co03917,MELO3Co20588,MELO3C008314,MELO3C025360,and MELO3C021658,were largely consistent wiht the transcriptomedata,suggesting their potentialrole inmelon waterlogging tolerance.Thesefindings provideafoundationforfurther explorationof waterlogging resistant genes in melon.
