Exploration of methods for extracting chondrocytes from autologous discarded cartilage tissue*

[Abstract]Objectie：Toinvestigateamethodfor extractingpurechondrocytesfromautologous discarded cartilage tissue. Methods：（1）Anoriginalcartilagecolectiondevicewasused tocolectdiscardedcartilagetissuefromcartilagedefectareas in knee jointcartilageinjurypatientsscheduledforautologouschondrocyte transplantation，folowedbychondrocyteextraction. （2）Cell morphology was observed under inverted phase contrast microscopy to determine conformity with chondrocyte morphology.Toluidineblue staining wasperformed toverify normal chondrocyte staining characteristics.Immunohistochemical staining was used to identify type collagen-secreting chondrocytes.Results： Cartilage mass： 72 mg; cell concentration： 1.736 |×| （20 （20 104 cels/mL.Primary celsappeared spherical，uniformly sized，suspended inculture medium with peripheral refractivity. After48hourculture，celsgrewinpolygonal shapeswithcolony-forming characteristics，consistentwithchondrocytegrowth paterns.Toluidine bluestainingshoweddark bluenuclei，light bluecytoplasm，and clearlyvisible contours，conforming to chondrocyte staining features.Immunohistochemical staining revealedyelow-brownnuclei，brown-yelow extracellularmatrix， andyellow-brownganulesaroundcels，indicating typeIcollagensecretion.Conclusion：Theoriginal“blade-filteraspirator" deviceenables eficient intraoperative recoveryofdiscarded cartilage.Thestepwiseenzymaticdigestion method sucesfuly extractshigh-puritychondrocytes，providinganovelcell sourceforautologouschondrocytetransplantationtechnologywhile avoiding iatrogenic injury caused by traditional harvesting.
