[Abstract]ObjectiveTo investigate the impactof transnasal administrationofferic ammonium citrate(FAC)on the sy naptic vesiclecycle pathway inolfactory bulb(OB)tissue,and toass theconsistencyof thisefect in PC12cells.Methods Mice were dividedintoFACtreatment groupandcontrolgroup.OBtissue wascolected,andRNA sequencing(RNA-seq)was performedtoidentifydiferentiallyexpressedgenesassociated withthesynapticvesiclecyclepathway;qPCRwasusedtovalidatethe changes in the expression of the genes such as Slc17a8 ,Slc6a12,and Uncl3c ,which was verified in PC12 cells. Results RNAseqshowedthatFACtreatment significantlydownregulatedtheexpresionof the genesasociated withthesynaptic vesiclecycle pathwayin OB tissue.The results of qPCR validation further confirmed significant downregulation of the Slcl7a8 ,Slc6a12,and Unc13c genes in both OB tissue τ=3.184-4.285 , P<0.05 )andPC12cells Δt=4.540-13.350 , P<0.05 ),which was consistent withthe resultsofRNA-seq.ConclusionIrontreatmentcandownregulatetheexpresionofthegenes associated withthe synaptic vesiclecyclepathwayinOBtisseandPC12cell,suggestingthatironmay impair thenormalfunctionofthesyapticvesicle cycle pathway.
