外源赤霉素对三青莴笋拔节影响及转录组初步分析

Abstract:To explore the mechanism of exogenous gibberelinon stem elongationof3-green(greenpeel,green flesh, greenleaf)asparagus letuce,twovarieties of3-gren asparagus letuce with diferent stem elongation ability,Tianxiangqing and Gaoshanqing,wereusedas experimental materials,andwater wassprayedascontrol(CK).Theeffectsof gibberellinon the growth of asparagusletuce were analyzed.Theresultsshowed thatcompared with theCK,the plant height of Tianxiang

qing and Gaoshanqing increased by 61.74% and 32.51% , respectively, andthe stem-to-leaf fresh weight ratio increased by 56.33% and 40.35% respectively,but there wasno significant effect on the fresh weight of aboveground partsper plant.GO analysis of differentially expressed genes showed that the differences between gibberellin treatmentand CK in the two asparagus lettuce varietieswere mainlyenriched in the biological process of cellular forma

tion,whilethediferencesbetweenthetwovarietiesweremainlyasociatedwithmuclearcomponentsandDNA-relatedbiologicalprocesses.Thediferentiallyexpresedgenesrelatedtostemelongationwerefoundtobeprimarilyenrichedinthediterpene biosynthesis pathway.Real-time fluorescencePCRvalidationshowedthatthe relativeexpresion levels of GA3oxgene in the shoot tips of Tianxiangqing and Gaoshanqing treated with gibberelin were 55.4% and 69.6% lower than those in the CK, respectively. The relative expression levels of GA20ox and KAO genes in Gaoshanqing asparagus letuce with good stem elongationability were higher than thosein Tianxiangqing asparagus letuce with weak stem elongationability.Insummary,exogenousgibberellincanafecttheexpressonofkeygenesintheendogenous gibberelinsynthesispathwayofasparagusletuce. The stem elongation ability of 3-green asparagus letuce is associated with the expression of LsGA20oxI (a critical gene involvedin endogenous gibberelln synthesis)and the transcription factor genes related to gibberellin metabolism.
