水稻抗甲咪唑烟酸突变体对多种ALS抑制剂类除草剂的抗性分析

Abstract:To create rice germplasm resistant to acetolactate synthase( ALS )inhibitorherbicides and promote the application of ALS inhibitor herbicides in rice production,ethyl methanesulfonate(EMS)mutagenesis and imazapic herbicidesprayingtreatment wereused toscrenrice mutantsresistanttoimazapic with Lianjing15astheexperimental material
inthis study.The mutation sitesand typesof the mutants weredeterminedbygeneticanalysisand genecloning. The wild-typeand mutantplantsweresprayedwithsterilewater (control,CK)and five commonlyused ALS inhibitor herbicides(imazamox,imazapic,imazethapyr,mesosulfuronmethyl and nicosulfuron)toanalyze theresistancelevel of themutant.The results showed that one rice mutant resist
antto imazapic was obtained.A nucleotide substitution from G to A was identified at position188O in the ALS gene, resulting in a serine-to-asparagine changeat the 627th amino acid residue in the ALS protein.The ALS activity,stem and leaffresh weight,rotfreshweightandtotalchlorophyll contentof wild-typeplants weresignificantlylowerthanthoseinthe sterile water control(CK)after spraying imidazolinone herbicides imazamox,imazapic and imazethapyr.In contrast,the mutant plants showed no significant diferences in these parameters compared to the CK.The ALS activity,stem and leaf fresh weight,rootfreshweightand total chlorophyllcontentofwild-typeandmutantplants weresignificantlylower than those of CK after spraying sulfonylureaherbicides mesosulfuron-methyland nicosulfuron.Therefore,the mutantsobtained in this study exhibit strong resistance to imidazolinone herbicides but weak resistance to sulfonylurea herbicides.
