Abstract:To investigate therole of juvenile hormone esterase(JHE)in the growth,development,and environmentaladaptationof Apocheimacinerarius,thisstudyscreenedand identifieda JHE genebased on the transcriptomic data of A.cinerarius viahomologous alignment,and named itAcinJHE.The CDSsequenceof this gene was1755bpin length, encodinga protein composed of 59O amino acids.The predicted relative molecular weight of the protein was 65580,with anisoelectric point of 5.70.It contained a signalpeptideof25aainlength,lackedtransmembrane structures,and itscarboxylesterase conserved domainwas locatedat39-579aa.Phylogeneticanalysisresultsshowed that AcinJHE had the closest genetic relationship with the JHEof Operophtera brumata,clustering on the same
branch.Fromthelarvalstagetothenon-diapausepupal stage,therelativeexpressionlevelofAcinJHEincreasedsignificantly ( P<0.05 ).Fromthediapause pupal stage to the diapause-terminated pupal stage,the relative expresson level of AcinJHE in A .cinerarius decreased significantly( P<0.05 ).Therelative expressionlevel of AcinJHE was the highest in the headsof male adults.The AcinJHE gene responded to low-temperatureand starvation stresss.Afterthreehours of low-temperature treatment at 4 C ,the relative expresson level of the AcinJHE gene in the 3rd-instar larvae of A.cinerarius reached thepeak.After72 hours of starvation treatment,the relative expressionlevel of the AcinJHE genein the4th-instarlarvae of A.cinerariuswas significantly higher than that in the control group ( P<0.05 ).This study elucidates the molecular mechanismoftheAcinJHEgene inregulating thegrowth,development,andstressadaptationofA.cinerarius,therebyprovidinga theoretical basis for its control.
