Abstract:The NAC transcription factors play important roles in plant growthand development as wellasin responses toabioticandbioticstresses.ToclarifythefunctionsofNACtranscriptionfactors in ViciafabaL.,inthis study, bioinformatic methods were used to identify the members of the NAC family genes from the Vicia faba genome database. The genestructures,cis-actingelements in promoters,chromosomal distributions,andthe physicochemical propertiesofthe proteinsencodedbythesegeneswereanalyzed.ByintegratingthetranscriptomedataofViciafabainfectedbyAlternariaalternata(thepathogen causingleaf spotin Viciafaba),candidategenesrelated todiseaseresistance werescreened.Theresults showedthatthere were77 membersintheNACtranscriptionfactorgene familyof Vicia faba,which wereunevenly distributed on six chromosomes.Theencoded proteinsranged insizefrom133aa to 665aa.The VfNAC proteinswere
dividedinto 16subfamilies,allofwhich contained the conserved NAM domain and werelocalized to the nucleus. The promoters of VfNAC genes contained various cis -acting elements such as light-responsive elements,hormone-responsive elements,and drought-responsive elements.The transcriptome sequencing results showed that 33 VfNAC genesweredifferentiallyexpressed in Vicia faba infected byAlternariaalternata,and four genesexhibiting significant differential expression were selected as keycandidate
disease-resistancegenes forfurtherstudy.Thisstudyprovidesareferenceforthesubsequentanalysisof thebiological functions of NAC transcription factors in Vicia fabaand theresearch on the molecular mechanisms of disease resistance.
