Research progress on the role of AIM2 like receptors inhost defense against bacterial and fungal infections

Abstract:The innate immune response serves as theprimary defense against pathogenic infections,with AIM2-like receptors(ALRs)representing pivotal components of the intracelular DNA-sensing receptor family.These receptors detect pathogen-derivedor host-derivedDNA,therebycontributing to intrinsicimmune defense mechanisms.Although theroleofALRsinantiviralimmunityhasben wellcharacterized,theirregulatorynetworksandfunctional heterogeneity inbacterialandfungalinfectionsremaininsuffcientlysummaized.Thisreviewintegatesrecentadvances tosystematically elucidatethe mechanisms ofAIM2and itsfamilymembersagainst bacterialand fungal pathogens.Speci ficall,AIM2 exertsanti-infective efects byinflammasome assmbly(recruiting ASCandactivating Caspase-1through pro-Caspase-1 binding),leading to pyroptosis and proinflammatorycytokine release,whereas other members like IFI16mediate broadspectrumantimicrobialactivityvia inflammasome-independentpathways,suchasdirectpathogen DNA binding to inhibit replication orcGAS-STING-mediated typeIinterferonresponses.These findingsdemonstrate that ALRscoordinately regulateanti-infectionimmunitythroughmultimodalmechanisms,andtheirpathogen-specificDNArecognitionpattrns offer novel perspectives for targeted therapeutic interventions against infectious diseases.
