Identification and Analysisof a Mixed Infection of Six Viruses in Sweet Potato Cultivar“Mashali"

AbstractTranscriptomesequencingandRT-PCRwereperformedonfourswetpotatosamplescolectedfromLinyiCityShandongProvince,tostudyttysofusttfeedswpotato“Mashl,noertrovidfercforhdigoffotato sedlingsandthefoulationofprevetionandcotroltrategies.Theseetpotatosamplesof“ashali”showdobviousosaicndcrikling symptoms.Sixknoviuses,incdingsweepotatofatheryotlevirus(PFV),swetpotatolatenvius(SPLV),sweepoatoeafcurl virus(SPLCV),sweetpotatovirusG(SPVG),sweetpotatovirusC(SPVC)andsweetpotatovirus2(SPV2)wereidentified.Thspecific primersoftheaboveviruseswereseletedforRPCRdetectio,tetargeandsiththeexpectedsizeofthesixviuseswereobaidand thetargetviussequences wereverifdbysequencing.TeresultsofRT-CRandtranscriptomesequencingwereconsistentwittesulsf 3samplescontaining6virusesand1samplecontaining4viruses.TephylogeneticanalysisbasedontheCPgeneshowedthattheetected SPFMV hadacloseevolutionalrelationshipwithSPFMVrusetcrack(RC),thedetectedSPLCV,SPV2andSPVCwere mostcloselyrelated totheisolatesofina,PVGasostoelyelatedtoholatesofCndtheeresiantfrecsogisolatefroV colectefrodftgiouloftsdyillprvdealsisforstablstofldotroldvfr nology system of sweet potato cultivar“Marasali”.
