Extractionand Antioxidant Activity of Total Flavonoids from Sea-buckthorn Branches and Stems

AbstractUsingsea-bucktobrancesasrmaterials，teetractionprocessoftotalfavonoidsasotiizedusinsinglefactorprmentsandresposesfaceethdology，nditsprotetieectolohlicliverjury（A）wssudiedTeesultssowdatetotal flavonoid content in the branches and trunks of sea-buckthorn was 0.64% . The optimal extraction process for total flavonoids from sea-buckthorn branches was ultrasonic frequency of 50kHz ，powerof 2OoW，extraction temperatureof 55∘C ，liquid-material ratio of 23：1，ethanol concentration of 61% ，extraction time of 2 hours.Under these conditions，the total flavonoid extraction amount was 4 .19mg/g . Total flavonoids were purified and enriched using X-5 macroporous resin. In vitro antioxidant tests showed that the 40% ethanol purified product had the best antioxidantactivity.ThescavengingratesofDPPH，ABTS，hydroxylandsuperoxideanionradicalswere3.17，37.O6，792.51and （204号 262.42μg/mL ，respectively.Cellexperimentsshowedthattotalflavonoids fromsea buckthornbranchescould effctivelyreduce thedamageof ethanol to HepG2 cells，and protect HepG2 cells through the Keapl/Nrf2/HO-1 pathway.
