University,Hefei,Anhui ) AbstractObjetie]ThereltioshipbetweenPCYP86A1,PCY86B1genesandpearplbrowntraitswasexplored.MethodThePbCYP86A1/B1genewasclonedfromthepelof“Dangshansu”pearand“Dangshanjinsu”pearbyreverse ranscriptionpolymerasechainreaction(RT-PCR).Thevirus-inducedgenesilencing(VIGS)methodwasusedtosilencethetargetgeneusingthetobacocrispycrackvirus (TRV)vector.Byanalyzing thecontrol group(TRV2-TRV1)andthe experimental group(TRV-PbCYP86A1,TRV-PbCYP86B1)pearpericarppheotyptomaticatiolatiexpressonoftgenedangsinompositioftesberin.Resultsuls showedthatafterTRV-PbCYP86AlandTV-PbCYP86Blwereinjectedintothepearepidemis,suberinintheinjectionpartscouldnotbe accumulatedwittfrutdvelopntadidihelowgreetcoldierebeweeneijeonsitendesurronger icarpwassignificantlygreatertantatoftecontrloup.-PCRidcatedthatthxpressosofYP86A1andPCYP86Bgeesre ducedsignfcantlywithsignifcantdierenceintheexocarof‘Xiusu’pearwithVIGScomparedihontrol.Conlusion]eresultsofsuberincontentanalysisshowdthatbothPCP86AlandPCYP86BgensilencingcouldsignificantlyihibithsynthesisofC6doy fatty acids and -diacid in pear pericarp,which will reduce the total amount of suberin.
