Abstract:3-bromo-2-oxocyclohexanecarboxamideasarawmaterialforthe synthesis of gliclazide is prepared from 2-oxocyclohexanecarboxamidevia bromination.Inorderto improvethesafetyand selectivityof brominationreaction, N -bromosuccinimide(NBS)wasused as a brominating agent for the bromination of 2-oxocyclohexanecarboxamide.Efects of stiring speed,reaction temperature,reaction solvent,NBS dosage and feeding method on the synthesis of 3-bromo-2-oxocyclohexanecarboxamide were investigated.The results show that the release of bromine from NBS and theconversion of the intermediate to 3-bromo-2-oxocyclohexanecarboxamide requires acid. Too much NBS and too fast addition of NBS would consume theacid quickly,which was not conduciveto theformation of 3- bromo-2-oxocyclohexanecarboxamide.With acetonitrileas solvent,NBS to2-oxocyclohexanecarboxamide molarratio 1.2:1,andNBS wasadded into thereactionsystem inthreebatchesatthe beginningof thereaction,2O minand 40 minrespectively,and the reaction was conducted forl05 min under reflux,then thecomplete conversion of 3- bromo-2-oxocyclohexanecarboxamidewasachieved,andtheyield of3-bromo-2-oxocyclohexanecarboxamide is 98% ·
