壳聚糖阳离子纳晶胶介质的制备及对乳铁蛋白的分离特性

Abstract:Anovel chitosan cation exchange nano-cryogel was developed to addressthelimitation of traditional lactoferrin separation methods from whey,which include complex separation process,low purity,high cost,and dificult in industrialization.Chitosanand polybutyl methacrylate nanogel were usedascomposite substrates,and cryogel embedded with nanogel (nano-cryogel)was prepared by cryo-polymerization,which was folowed bygrafting with2-acrylamido-2-methylpropane sulfonicacid to obtain thechitosan-polybutyl methacrylatenano-cryogel with cation exchange function. The medium was employed for lactoferrin separation under conditions of loading whey at pH 5.8,diluting twice with buffer at a flow rate of 1cm⋅min-1 ,and then eluted with NaCl solution at concentrations of 0.1, 0.3 and 1.0mol⋅L-1 gradiently. The results demonstrate that 0.3mol⋅L-1 of NaCl solution elutes most of the impurity proteins,while 1.0mol⋅L-1 of NaCl solution elutes high purity of lactoferrin with corresponding purity and recovery rate of 92.7% and 86.3% ,respectively. These findings indicate that the nano-cryogel medium has significant potential for lactoferrinchromatographicseparation.

乳铁蛋白(lactoferrin)是一种由700多个氨基酸构成的多肽链,其分子质量约为 80kDa ,等电点约为8.5,广泛分布于哺乳动物组织中[1],可调节骨髓细胞生成,极大地促进细胞生长[2],且具有广谱抗菌、抗病毒、抗氧化能力[3-4]。(剩余11726字)
