ABSTRACT:This studyaimed tooptimize theultrasonic-assted enzymatic extraction processof Inonotus obliquus poly saccharides and evaluate their inhibitory activity against α glucosidase. Single-factor experiments were conducted to identify key factors afecting the polysaccharide extractionrate,including enzyme dosage,liquid-to-solidratio,ultrasonic temperature andultrasonic time.Orthogonal tests were then used tooptimize theextractionconditions,which revealed that the optimal conditions were an enzyme dosage of 1% ,a solid-to-liquid ratio of 1:20 ,an ultrasonic temperature of 50∘C and an ultrasonic time of 20min .Under these conditions,the extraction rate of I .obliquus polysaccharides reached 4.97% . Furthermore, I.obliquus polysaccharides demonstrated significant inhibitory effects on α glucosidase,with an IC50 value of 0.296mg⋅mL-1 ,indicating a potent α glucosidase inhibitory activity and its potential application in blood sugar-lowering therapies.
