Abstract:In order to develop a credible dual fluorescence quantitative polymerase chain reaction (qPCR) for detecting novel goose astrovirus type1(NGAstV-1)and type2(NGAstV-2),primers were designed to establish a dual fluorescenceqPCR detection method using ORF2 proteinof NGAstV-1and NGAstV-2,and36samplesof goose liverand kidneytisuesuspectedtobeinfectedwithNGAstV-1andNGAstV-2 were evaluated inthis study.The test results showed thatthe correlation coeffcients of qPCR were O.997and were negative for novel duck reovirus (NDRV),avian influenza virus (AIV),fowl adenovirus(FAdV),and goose parvovirus(GPV).The minimum detection limit was 1.12×101copies/μL .Thereproducibility test results indicate good reproducibility,witha coefficient of variation between groups of less than 0.8% andacoefficient of variationwithin groups of less than 0.5% It canbeseenthattheqPCRcan eficientlyandspecificalldetect NGAstV-1and NGAstV-2,providingafeasible method for NGAstV monitoring.
