体细胞核移植技术在蓝塘猪遗传资源保护中的应用

Abstract：This study aimed to clone theLantang pig using somaticcellnuclear transfer technology，offring a noveland eficient approach for preserving thegenetic resources of exceptional local and endangered pig breeds in China.We compared the tisue block method and collagenase digestion method to establish Lantang pig somatic cell lines.Additionaly，we analyzed the developmentaloutcomes of cloned embryos created from Lantang pig donor cells of diffrent genders in vitroand transplanted them in vivo.Theresults indicated thatthecollagenase method yielded cells more rapidlyandin higher quantitiescompared tothe tissue block method.While there were no notable differences in cleavagerates and total blastocystcellcounts between cloned embryosof diferent genders in vitro，the blastocyst rate of male embryos was significantly higher than that of female embryos（ P<0.05 ）. Through in vivo transfer of clonedembryos from Lantang pigs in threeseparate batches，wesuccessfullyobtained four healthycloned piglets. This study marks the firstsuccessul caseof somaticcellcloning of theLantang pig in China，providing adependable technical tool for preserving the genetic resources of this breed.
