[Abstract]ObjectiveToprepareaphotothermal therapeuticmicronedleforthe treatmentof age-related chloasmathatinhibitsmelaninsecretion,andtoevaluateitsperformanceandeficacyMethodsGAGOnanocompositeswerepreparedbymodifying thegrapeneoxide(GO)structure withgalicacid(GA);thesynthesisofGAGOwasverifiedusing UV-Visspectrophotometryand Fouriertransform infraredspectroscopy;thestabityofGOandGAGO inaqueoussolution,PBSandRPM1640 medium wasdetected usingaMalvemparticlesizer;three matrix materials,hitosan,poly-vinypyolidone(PVP),andpolyinylalcohol,wereeed andthe micromolding perfusionmethodwasdetermined.Their topographywascharacterizedusing ightmicroscopyandscaning electronmicroscoy;mechanicalpropertiesofmicrondlesweretestedusinganIstrostroncompresio-tesiontester;skinsertion propertiesofmicroneedlesweretestedusing SDratskin;photothermalconversionpropertiesof microneedleswereasessedbyan 808nmnear-infrared(NIR)laser;andtheinvitroinhibitoryefectofGAGOphotothermal therapywasverifiedusingB16melanoma cels.ResultsGAwassuccessullyloadedonGOandhad goodstabilityandphysicochemicalproperties.GAGO-PVPmicroeedles weresuesfullyconstructed,withbeautifulappearance,easypouringanddemoulding,intactneedetipswithoutbubbles,and throughmechanicalpropertistesting,telossofficacyreached3.66Nneede,hichanpeneatetesinwitoutcausigla mation,andhasgoodphototheralconversionproperties.Comparedwiththecontrol group,GAGOcouldsignificantlyincreasetheac tivityofB16melanomacell,ecreasetyrosinaseactivityandinbitmelaninseretionduringNRchallnge,andtedierecsere statistically significant( P<0.001 ).Conclusion GAGO NIR treatment significantly inhibited B16 melanoma cell viability and melanin secretion,and GAGO-PVP microneedles have the potential to be applied in melasma treatment.
