补肾宁神膏的指纹图谱建立、含量测定及体外抗氧化作用的谱效关系研究

ABSTRACTOBJECTIVEToestablish thefingerprint of Bushenningshenointment,determinethecontentsof its major constituents,andinvestigateitsinvitroantioxidantactivity.METHODsHighperformanceliquidchromatography(HPLC) fingerprintsof1batchesofBushen nngshen ointment were established.Similarityevaluationand identificationofcommonpeaks weresubsequently performed.Thecontentsof1Ocomponentssuchassalidrosidewere determinedusing thesame HPLCmethod. Using the scavenging rates against 2, 2′ -azino bis (3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)and1,1-diphenyl-2- picrylhydrazyl(DPH)adicals,aswellasfericionreducingantioxidantpower(FRAP)asindcators,theanti-oxidantactivityof theointment was evaluated;greyrelationalanalysisandpartialleastsquaresregresion wereconductedusingSMCA14.1software toestablishthespectrum-efectrelationship.RESULTSThefingerprintchromatogramof10batchesof Bushenningshenointment contained 24 common peaks, with similarity valuesallexceeding O.96.Eleven peaks were identified as adenosine(peak1),salidroside(peak4),morroniside(peak6),catechin(peak7),paeoniflorin(peak 1O),spinosin(peak11),ferulic acid (peak 12),isoquercitrin (peak 13),verbascoside(peak 14),paeonol (peak 23),and emodin(peak 24).Content determination results showed that theaverage contentsof salidroside,morroniside,catechin,paeoniflorin,spinosin,ferulicacid,isoquercitrin,verbascoside,paeonol,andemodinwere0.725,1.962,0.214,3.395,0.124,0107,0.286, 0.019,0.034 and 0.067mg/g ,respectively.The antioxidant potency composite index(APC)for the 10 batches ranged from 85.08% to 96.35% .Spectrum-effect relationshipanalysis indicated that all 24common peaks were positively correlated with the antioxidant capacity. Seventeen peaks had variable importance in projection values >1 ,specitically peaks 2,5,6,7,9,10,13- 21,23,and24.CONCLUSIONSThis studysucesfullyestablishedthe HPLC fingerprintandcontentdetermination method for Bushenningshenointment.Thecompoundsrepresented bythe17commonpeakssuch as moronisidemaybetheactivecomponents contributing to its antioxidant effects.
