Analysis of The Chemical Composition of Rougan Granules Based on UPLC-Q-TOF-MS Technology

Abstract：ObjectiveToanalyzeandidentifythechemicalcompositionof Rougan Granulesandtoelucidatetheposiblecleavage pathways of someof hecompounds containedtherein.Methods UtraPerformanceLiquid ChromatographyTandemQuadrupole Timeof-flight Mass Spectrometry（UPLC-Q-TOF-MS）wasusedtoanalyzeandidentifythechemicalcompositionof RouganGranules， and the chromatographic column was an Agilent C18 reversed-phase chromatographic column （ 2.1mm×100mm， 1.8μm） ，with the mobile phases of 0.1% formic acid in water A-acetonitrile B in a gradient elution，and the flow rate of 0.3mL/min . Scanning was performed in positive and negative ion mode with a scanning range of m/z50-1 500.Based on the secondary fragmentationioninformation，techemicalcompositionsintheRouganGranuleswereidentifidincombinationwithliteraturedata.ResultsAtotalof82 compoundswereidentified，mainlyterpenoids，flavonoids，phthalides，phenylpropanoids，glycosidesandalkaloids.ConclusionThe establishedmethodcansystematicalyqualitativelyanalyzethchemicalconstuentsinRougnGranulesndprovideatheoreticalbsis for the study of Rougan Granules.

柔肝颗粒是江苏省中医院研制，用于治疗肝硬化的院内制剂（苏药制字Z04000542），由当归、赤芍、三七、泽兰、女贞子、牛膝、黄芪、白术、防己等9味中药组成，其中当归、赤芍养血柔肝为君药；泽兰、三七活血化瘀，牛膝、女贞子滋肾养肝为臣药；黄芪、白术、防己补土荣木，健脾利水，为佐药，诸药并用，具有柔肝健脾、活血通络之效。（剩余12436字）
