Analysis of The Chemical Composition of Rougan Granules Based on UPLC-Q-TOF-MS Technology

Abstract:ObjectiveToanalyzeandidentifythechemicalcompositionof Rougan Granulesandtoelucidatetheposiblecleavage pathways of someof hecompounds containedtherein.Methods UtraPerformanceLiquid ChromatographyTandemQuadrupole Timeof-flight Mass Spectrometry(UPLC-Q-TOF-MS)wasusedtoanalyzeandidentifythechemicalcompositionof RouganGranules, and the chromatographic column was an Agilent C18 reversed-phase chromatographic column ( 2.1mm×100mm, 1.8μm) ,with the mobile phases of 0.1% formic acid in water A-acetonitrile B in a gradient elution,and the flow rate of 0.3mL/min . Scanning was performed in positive and negative ion mode with a scanning range of m/z50-1 500.Based on the secondary fragmentationioninformation,techemicalcompositionsintheRouganGranuleswereidentifidincombinationwithliteraturedata.ResultsAtotalof82 compoundswereidentified,mainlyterpenoids,flavonoids,phthalides,phenylpropanoids,glycosidesandalkaloids.ConclusionThe establishedmethodcansystematicalyqualitativelyanalyzethchemicalconstuentsinRougnGranulesndprovideatheoreticalbsis for the study of Rougan Granules.

柔肝颗粒是江苏省中医院研制,用于治疗肝硬化的院内制剂(苏药制字Z04000542),由当归、赤芍、三七、泽兰、女贞子、牛膝、黄芪、白术、防己等9味中药组成,其中当归、赤芍养血柔肝为君药;泽兰、三七活血化瘀,牛膝、女贞子滋肾养肝为臣药;黄芪、白术、防己补土荣木,健脾利水,为佐药,诸药并用,具有柔肝健脾、活血通络之效。(剩余12436字)
