Construction and cell imaging of GSH-responsive curcumin probe

[Abstract]ObjectiveTosynthesize a glutathione (GSH)-responsivecurcumin-based fluorescent probe (CPB)andapply it forhepatocellarcarcinomacellimaging,investigatingitscytotoxcitymitochondria-argetingcapability,andabilitytoinduce reactiveoxygen species (ROS)generation Methods A GSH-responsive fluorescent probe (CPB)was synthesized using curcuminasa fluorescentmoietyanditsstructurewascharacterizedbyliquidchromatographyassspectrometry(C-MS)anduclearmagnetic resonance (NMR).Ultraviolet (UV)spectroscopywasused toanalyzeitsresponsetoGSHandtimedependentchanges.Thecytotoxicity ofCPBwasaessdbsaynditsgetielodioiaonicellagingdhodalcalatio experiment.Results Theresultsof NMRcarbon spectraandLC-MSconfirmed thesuccesful synthesisof CPB.UVspectra showedthattheresponsesignalofCPBitself wasconcentration-dependent,whilethatofCPBtoGSHwastime-dependent.MTT assay showed that CPB of 20-30μmol/mL significantly inhibited the proliferation of HepG2 cells,and CPB of 40μmol/mL (20 inhibitedtheproliferationofBEL7402/DPcells,whileshowingalmostnotoxicitytoAML-12cels,indicatinglowcytotoxicity.ROS assay showed that CPBinducedROS generationin HepG2andBEL7402/DDPcelsinaconcentration-dependent manner. Comparedwiththecontrol group,thecellimaging experiment demonstrated that thefluorescence intensityof CPBenhancedat concentrationsof10\~3Oμmol/Landitsfluorescencesignalslargelyoverlappedwiththoseofcommercialmitochondrialprobes. Conclusion Inthispaper,CPB,aGSH-responsivecurcumin-basedfluorescentprobe,wasdesignedandsynthesized.Itexhibits goodGSHresponsesensitivitylowcytotoxicity,andmitochondrialtargeting,whichcanbeappliedasasafeandreliable GSH detection probe for tumor cell imaging.
