Molecular Identification Analysis of Pre-harvest Sprouting Resistance Traits in 194 Wheat Varieties (Lines)

Abstract: Pre-harvest sprouting (PHS)can lead to reduced wheat yield and deteriorated grainquality,posing a serious threatto wheat production.Identifying thePHSresistance of planted wheat varieties and promotingthecultivationof those varieties pyramidedwithPHS-resistant genes isoneof the most economical approaches tomitigate the damage caused by pre-harvest sprouting.This study evaluates 194 wheat varieties (lines)from the Huanghuai Wheat Region, using the seed germination method to determine their germination index (GI),thereby clarifying their PHS resistance. Resultsshow continuous variation in GI values (5%-86%) among the tested materials,indicating significant differences in PHS resistance.Among them,there is1 variety (line)with high PHS resistance,23 with moderate PHS resistance,97with moderate PHSsusceptibility,and73 with high PHSsusceptibility.The PHS-susceptible materials amount to 170,accounting for 87.63% .Additionally,molecular markers are used to detect previously reported PHSrelated genes (TaVp-1,TaPM19-A1,TaDFR,TaQSD1,TaMKK3-A,TaMFT-3A,etc.).Theresults show that TaDFR and TaQSDl significantly influence PHS resistance of the tested wheat varieties (lines).Byscreening based on PHS resistance,a total of9 germplasm resources are identified: Zhengmai 113,21P85,Zhongyu 149,Zhumai 1011, Zhongpinmai121,Yuzhouzimai6,Yuzhouzimai7,Jiamai17,and Xinglanmai 99.These materialscontain both QD1 andDFR-Bgenesand exhibit moderateorhigherPHSresistance.Thisstudyclarifies thePHSresistanceand the distribution of PHS resistance genes among the 194 wheat varieties (lines),and screens out excelent germplasm resourcescarrying PHSresistancegenes,providing theoretical referencesand superiorgermplasm for thegenetic improvement ofwheatPHS resistance.
