Abstract:The Bacillus thuringiensis (Bt)gene,animportant exogenous gene ininsect-resistant transgeniccrops,playsa crucial role in crop pest control.The cultivation and promotion of Bt transgenic crops have greatly reduced the use of synthetic insecticides and brought huge benefits to farmers.However, the safety of Bt transgenic crops has gradually atracted people'sattention.Therefore,itisofgreatsignificance todevelopasimpleandsensitivemethodfordetectionof Bt transgene in crops.This study constructs a dual-modified triple helix DNA fluorescent molecularswitch (DTHFMS)and systematicall investigates the influencing factors and variation rules of the stabilityof DTHFMS.Theresults show hat DTHFMSwithalternatingtripletsofT-AoTandC-GoCdemonstrates properstabilitymakingitsuitableforhighlysensitive detection of Bt transgenes in maize seeds.This study achievesexcellent detection results by optimizing key detection parameters. The limit of detection is 0.05pM ,with a linear range of (0.1-6.0×104)pM :
