Theapplicationof Danio reriomodel in developmental toxicologyresearch

AbstractDanio rerio exhibits characteristics such as rapiddevelopment,external fertilization,strong reproductive capacity,and overalltransparencyduring earlydevelopmental stages,making them widelyusedas model organisms in developmentaltoxicologyresearch.TheapplicationofDanio rerio inthe safetybioactivityof traditional Chinese medicines (TCMs)andthetoxicityassessmentof environmentalpolltants weresummarizedfrom3dimensions:cardiacdevelopmental toxicity,neurodevelopmentaloxicityandvascularandbloodcelldvelopmentaltoxicityIersofrdiacevelotal toxicity research,atspecific doses,traditional Chinese mediciecomponentssuchastotal platycodinandbrucinecan induce bradycardiainzebrafish,accompaniedbysymptomsincludingcardiacarrstandhemorrhage.Environmentalpollutats like polybrominateddiphenylethers,polychlorinated biphenyls,andperfluorooctanoicacidcan triggercardiac oxidativestress in zebrafish,leading tocardiacdevelopmentaldefects.Regardingneurodevelopmentaltoxicityatspecificdoses,substances such ascadmiumchloride and bisphenolFexert neurotoxicitybyinterfering withthe expresionofkey genes involved in neural development.Regarding vascularand bloodcelldevelopmental toxicitystudies,relevantresearch hasshownthatsome TCM extracts can inhibit (e.g., methanol extractof Xihuang Wan)or promote (e.g.,total saponins from Panax notoginseng roots andflowers)Danio rerio angiogenesis,whileothers havenosignificant impact (e.g.,waterdecoctionof Geko gecko). Environmental polltants such as mycophenolic acid (MPA)can inhibit Danio rerio vascular development byregulating specific receptors (e.g.,AHR)or through direct effects.This review providesa reference forthe subsequentresearch and practical application of Danio rerio.
