超声处理对泡核桃雄花总多酚提取效果的影响及其特性

ABSTRACT:The Juglans sigillta Dode male flower extracts can reduce theoxidative stressand inhibit theoxidationof macromolecules.In this study,onthe basis ofoptimizing the beter extraction conditions of male flower total polyphenols,its components andantioxidant efect invitro were investigated,andtheultrasound mechanism was preliminarily described.Total polyphenols extraction conditions of male flowers was optimizedby multiplequadratic regression equation;its components were analyzed by HPLC;Theantioxidant activityof total polyphenol was evaluated bydetermining theclearance of superoxide anionand hydroxylradical;The microstructureof walnut male flowers powders before andafter ultrasound treatment was observed by SEM. The results showed that the ultrasound power was determined at 250W . the beter extraction process of total polyphenol of walnut male flowers was as follows: solid to liquid ratio 1:60 ,treatment time 21min ,ethanol concentration 50% ,treatment temperature 61°C ,the total polyphenol yield of walnut male flowers was (24.45±2.12)mg⋅g-1 ; Four kinds of monomeric phenol were identified by HPLC from walnut male flowers.Among them,chlorogenic acid and rutinum were higher,and gallic acid and quercetin were lower;total polyphenol, vitamin C(Vc)and BHTcould remove superoxide anions and hydroxyl radicals.In terms of its clearancecapability, Vc> total polyphenol > BHT;ultrasound treatment could tear and destroy the tissue surface of male flower,and release more plant polyphenoland strengthen theextractionefect.The resultscan provide the possibilityforits applicationof male flowers polyphenols in the field of food storage and preservation.
