CACNAIC 和 RYR2 基因核酸多态性与蛋壳品质的关系

ABSTRACT:The up-regulation of CACNAlC(CaV1) expression in the calcium regulation pathway inhibits the expression ofryanodine receptor (RYR2)gene and promotes calcium ion transport and eggshellformation.This study analyzed the polymorphism of CACNAiC and RYR2 gene promoter region and its relationship with eggshell quality of Wuhua chicken. Total DNA was extracted from wing vein blood of 340 35-week-old Wuhua chickens with complete egg production records. PCR amplification and sequencing were used to screen SNPs in the promoter regions of CACNAlC and RYR2 genes.Eggs were collcted for 3 days for eggshellquality detection,and5 chickens were randomly slaughtered to take heart,liver,spleen,lung,kidney,chest muscle,leg muscle,ovary,enlarged part,uterus,isthmus and other tissues for gene expression quantification. Theoverexpressionand knockdown plasmids of CACNA1C gene were constructed and transfected into DF-1 cells to analyze their regulatory effects on the expression of RYR2 gene. The results showed that one SNP site was screened at -2 000 bp in the promoter region of CACNA1C and RYR2 genes, respectively. CACNA1C gene g.61191372C>G locus produced three genotypes. The eggshell strength of CC was significantly higher than that of CG ( P<0.05 .Theg locus of RYR2 gene produced three genotypes. The eggshell thickness of AG and AA was significantly higher than that of GG ( P>0.05 ). The expression of CACNAlC in the enlarged part wassignificantly higher than thatin the uterus,ovary,isthmus,heart,liver,splen,lung,kidney,chest muscle and leg muscle. Overexpression of CACNAlC gene CC type inhibited RYR2 expression. In conclusion,the CC type of CACNAlC gene promoter region can reduce the expression of RXR2 gene,promote calcium ion transport, increase eggshell density,and improve eggshell strength.
