ABSTRACT:To generate a thermostable and combined vaccine candidate against the infectious laryngotracheitis(ILT) and Newcastle disease(ND),this study cloned the gB gene of ILTV to the thermostable NDV NDV/rAHRo9 strain using the seamlesscloning technique.Then,the Western blot experiment was used tocheck the expresionof gB,and the thermostableand virulenceassssment experiments wereused todetectthebiologicalcharacteristicsof therecombinantvirus. The results showed that the gB gene of ILTV was successfully inserted into the pNDV/rAHRo9,and the rescued recombinant virus showeda high haemagglutination (HA)titer.Aditionaly,the Western blot assay confirmed the expression of the gB protein. The thermostable assessment showed that the recombinant viruses maintained a high HA activity and virus titer after exposure to 56∘C for about 6O minutes. Pathogenicity assessment data indicated that the recombinant virus hada mean death time(MDT)in chicken embryos of148 hours,an intracerebral pathogenicity index (ICPI) of O,andan intravenous pathogenicity index (IVPI)ofO.1,suggestingthat therecombinantvirus possssd typicalcharacteristics of anatenuated NDV.Inconclusion,the thermostable vacine candidatefor ILTand NDwas sucefullydeveloped in this study,which provides a novel approach for developing a live vector vaccine against ILTV.
