Abstract:Callus induction is crucial for successful genetic transformation,while WOX genes might enhance this processby stimulating celldivision shoot organogenesis.This study systematicallyanalyzed the WOX gene family in common bean(PhaseolusvulgarisL.),resulting in identification 18WOX genes that encode proteins with177 to 847 amino acids. These proteins are predicted containing homeodomain conserved domains to be localized in thecell nucleus.The promoterswere identified containing cis-acting elements related to meristem formation auxin response.Acolinearity phylogenetic tree analysis was conducted with the members the WOX gene family in mung bean, soybean Arabidopsis thaliana. The 124WOX genes can be divided into three major groups, the WOX homologous genes among the four species are subject to purifying selection. Through screening the media with different concentrations 6-BA 2,4-D hormone ratios,the callus induction systems two common bean varieties were successfully established.Through transcriptome combined with qRT-PCR analysis,eight diferentially expressed WOX genes were identified. The PvWOX1,PvWOX15 PvWOX16 genes were significantly corelated with thecallus inductionrate, their transcriptional trends were further confirmed using qRT-PCR. These results indicate that the PνWOXI , PvWOX15 PνWOXI6 genes can be used as potential target genes for improving the genetic transformation efficiency in common bean.
