Abstract：To investigate theregulatory mechanismof gene expresion inEriocheirsinensis（Chinese mittencrab）upon infection with Aeromonas hydrophila，wecomparedthe diferential gene expression profilesofhepatopancreas tissues between maleand femaleindividualsat24hourspost-pathogeninfectionthroughRNA-Seq，andpreliminarilyanalyzedwhetherhereis immunedimorphismbetweenmaleandfemaleEriocheirsinensis.Theresultsshowedthatatotalof3447diffrentiallyexpressed genes （DEGs）were identified between the infected groupand thecontrol group infemalecrabs，including1424upregulated genes and2O23 down-regulated genes.In malecrabs，3O35DEGs were enrichedbetween the infected group and thecontrol group，with1462up-regulated genesand1573down-regulated genes.The immune-relatedDEGsenriched inboth femaleand malecrabs mainlyincludedubiquitin-protein ligasegenes，Tol-likereceptor genes，C-type lectingenes，heat shock protein genes，andlysozyme genes.GO enrichment analysis revealedthata large number of DEGs wereenriched in immune-related biological processes.KEGGenrichmentanalysisindicated thatDEGsweresignificantlyenrichedin118pathways，and theimmune-related metabolicpathwaysmainlyincluded lysosome，extracelular matrix（ECM）-receptorinteraction，peroxidase，and phagosome.Inaddition，nine immune-relatedgenes fromthetranscriptomedatawereselectedforqRT PCRvalidation.Theresultswere consistentwith thosefrom RNA-Seq.This study preliminarilyexplored the molecular mechanism underlying the hepatopancreas of Eriocheir sinensisin response to Aeromonashydrophila infection.
