Abstract:Inordertodevelopahigh-performance liquidchromatography-masspectrometry(HPLC-MS)methodforquantitatively detectingendogenouslowconcentrationsofN-acylhomoserinelactones(AHLs)inbacteria,thisstudyextractedAHLsproducedby bacteriathoughquidlqdetractiotiedastroetryndomatogapicoditio,ndtablshddiote withgoodlinearelatioshipallatrixect,ndigecovryate.Tultsodatteearelationoeints(f the seven AHLs were all greater than O.995,and the r valuesof all AHLs were greater than O.99o,indicating that the method hasa good linearrelationship.The substrate effectsof8 typesofAHLsare all greater than 80.0% ,andthebacterial growthmatrixhasarelatively smallimpactonthdetectionofendogenousAHLs.ThestudyalsofoundthatAHLswithlongercarbonchainshavelessinteferencein complexmatrices,ilesotaiingcaboylgrouseibitacertainsigalsuppressfctTheecoveryatesof7tysofHs all exceeded 80.0% ,indicating that this method has highrecoveryrates and goodaccuracyandreliability.The method was used todetect endogenousAHLsin bacteria,and theresults showed that C 10⋅ -HSLand C 12 -HSL were themain AHLs,furtherverifying the effectiveness of the method.Thismethodcanaccuratelydetectendogenous traceAHLsinbacteria,providingreliabletechnicalsupportforstudying endogenous AHLsin bacterial quorum sensing(QS)systems.
