Abstract:This study established a systematic approach for characterizing theanti-inflammatorycomponents of the classic prescription Huaihua Powder using ofline two-dimensional liquid chromatography coupled with QOrbitrap mass spectrometry (2D-LC-Q-Orbitrap MS). First, the extract of Huaihua Powder was fractionated and enriched based on relative retention time by reversed-phase ultra-high performance liquid chromatography in the first dimension. The inhibition of nitric oxide (NO) production in LPS-induced RAW264.7 macrophages was used as an in vitro evaluation index to screen for anti-inflammatory active fractions.The results revealed thatthree fractions (A,B,and C) exhibited significant anti-inflammatory activity.Furthermore, guided by this anti-inflammatory activity,the chemical constituents of these active fractions were identified in the second dimension using UPLC-Q-Orbitrap HRMS technology with a combination of hydrophilic and reversed-phase columns.A total of 106 compounds were identified.Fraction A primarily contained amino acids,carboxylic acids,and alkaloids;Fraction B was dominated by flavonoids; and Fraction C also contained a considerable number of flavonoids.By integrating efficient separation, activity evaluation,and chemical characterization, this method enables rapid and comprehensive identification of the anti-inflammatory active components in Huaihua Powder,providing a reliable technical foundation for further research on its pharmacodynamic material basis.
