AbstractIn recent years, duck adenovirus type 3(DAdV-3) has been reported in multiple regions,resulting in substantial losses to China’s poultry industry.The fiber protein (Fiber)plays acritical role in mediating DAdV-3 infection and inducing humoral immunity.In the present study,the Fiber-1 protein of DAdV-3 was recombinantly expressed using the insect baculovirus flashBAC expression system to obtain the target protein. Owing to the integration of the LacZ gene into Autographa californica multiple nucleopolyhedrovirus (AcMNPV) in this system,LacZ expression was initiated post-recombination,enabling the screeing of positive viruses via blue-white selection.This approach obviated the need for plaque screening,thereby considerably reducing the experimental time.Westem blot analysis validated the successful expression of Fiber-1 protein in Sf9 insect cels. Theresultsof this studycould laya foundation for further investigating therole of Fiber-1 protein in the infection mechanism of DAdV-3 and for the development of related biological products.
