Optimization of Rice Genetic Transformation Mediated by Agrobacterium Tumefaciens

Abstract:Objective:TooptimizethegenetictransformationofricemediatedbyAgrobacterium.Method:Mature embryos ofindicaricevariety9311,Minghui-86,andjaponicaricevarietyNihon Harunawereusedasresearch materialsfor genetictransformationoptimizationofrice.TheAgrobacterium strainusedintheexperiment was LBA404,and the plasmid used waspCAMBIA1301A At MYB44.After preparing multiple solutions,prepare Agrobacterium competentcells. Transfer plasmid pCAMBIA1301A At MYB44 into Agrobacterium strain LBA4404 using prepared Agrobacterium competent cells.Applytheimportedbacterialsolutiontoinfectthreetypesofriceandachievetheirgenetictransformation.Extract DNA molecules fromrice plantsafter genetictransformationandperformPCRdetection.Result:Afteroptimization,the DNAextractionamount,DNApurity,andDNAintegritywereallhigher.Afteroptimization,thegenetictransformation efectofindicaricevarietiesisbeter.Afteroptimization,thePCRbandsareclearer,andtheocurenceofnon-specific bandsis lessobvious,resulting inahigher positiverateofPCRdetection.Conclusion:TheoptimizationofAgrobacterium mediatedrice genetic transformationhas beensuccesfullyachievedinthestudy,which hassignificantpractical significance.
