Abstract:Objective:To predictthe mechanismof the Sophoradavidi flower extract(SDFE)in preventing and treating pulmonary nodules based on network pharmacology.Methods:The SwissTarget Predictionand GeneCards databases wereusedtoobtainthetarget genes ofcompounds in SDFEandthe therapeutic targetsfor pulmonarynodules,respectively, follwed byidentifying their intersection targets.The STRINGonline platform was employed toconstructaprotein-protein interaction (PPI) networkof theintersectiontargets.GOand KEGG enrichmentanalyses were performedontheintersection targetsusing theDAVID database.Acompound-intersection target-KEGG networkwas constructed using Cytoscape software,and molecular docking validation was conducted using Discovery Studio software.Results:92 intersection targetswere determined.GO functional enrichment analysis revealed thatthese targetswere primarily associatedwith transmembrane receptor protein tyrosine kinaseactivity,positive regulationof kinaseactivity,andreceptorcomplexes. KEGG enrichment analysis indicated significant involvement in pathways in cancer and PI3K-Akt signaling pathway.The commoncore targets inboth thePPI networkandthecompound-intersectiontarget-KEGG network were EGFR,MMP9, SRC and AKT1.The top10core compounds were palmatine,medicarpin,genkwanin,pinocembrin,7-methoxyluteolin, nepetalactone,naringenin,diosmetin,luteolinand matrine.Moleculardocking results showedthatluteolinandmatrine hadLibDockScores greater than10O whenbinding to core targets such as EGFR,MMP9 and SRC.Conclusion:SDFE canprevent and treatpulmonary nodules through multiplecomponents,multiple targets and multiplepathways,providing a theoretical basis for subsequent experimental validation and drug development.
