NSE、SAA水平与高胆红素血症新生大鼠脑损伤的相关性分析

ABSTRACT:The corelation between neuron-specific enolase (NSE)and serum amyloid A(SAA)levels and brain injury in neonatalrats with hyperbilirubinemia was analyzed. Control group and model group were setup with 4 newborn rats in each group.The brain tissue of rats was observed by staining.The level of serum bilirubin was measured by microplate method.The mRNA and protein expressions of NSE,SAA,Bcl-2 and Bax were detected by PCR and Western blot. Prismsoftware wasused toanalyze thecorelation betweenNSEand SAAlevelsand brain injury inneonatalrats with hyperbilirubinemia.The results showed that:The hyperbilirubinemic neonatal rat model was successullyconstructed;The mRNA and protein levels of NSE and SAA in brain tisseof model group were significantly higher than those of control group;The levels of Bcl-2 mRNA and protein in the brain of the model group were significantly lower than those of the control group,whilethelevelsof Bax mRNAand protein were significantly higher than thoseof thecontrol group;Corelation analysis showed that both NSE and SAA were negatively correlated with the expression ofBcl-2,while both NSE and SAA were positivelycorrelated with the expression of Bax.The expressions of NSE,SAA and Bax in the brain tissue of hyperbilirubinemiarats showed an increasing trend,while the expression of Bcl-2 was decreased.The changes in the expresion of Baxand Bcl-2canaffct the severityofbrain injury. Therefore,the expressionofNSE,SAAcanbe regulated clinicall throughdrugor biological means,which mayinhibit inflammationand neuronal injuryand reducetherisk of brain damage.Inthefuture,it is necessarytofurther verifyits therapeuticfeasibilityand promote the aplicationof the integrated strategy of diagnosis and intervention.

高胆红素血症是临床上常见病症,多以新生儿为主要发病人群,多数患儿症状较轻且具有自限性,少数患儿病情严重,通常以高胆红素脑病、神经损害以及遗留神经系统后遗症为主要症状,严重者会危及生命[1-3]。(剩余10607字)
