艾地苯醌对湖羊精液低温保存效果的影响

ABSTRACT：The study aims to investigate the efect of adding Idebenone （IDE）to the semen extender on low-temperature preservationof semen，and to provideatheoreticalbasis foroptimizing the semen extenderandimproving thequality of semen after preservation. In this study，fresh semen was collected from 5Hu sheep aged 2-3 years old with similar body weights and good healthconditions.The semen that passed the test was mixedand diluted with diferent concentration of IDE before being stored atlow temperature.Thequalified semen was mixed and diluted with diluents containing different concentrations of IDE [O group （control group）， 5μmol⋅L-1 group， 10μmol⋅L-1 group and （204 group] for cryopreservation，with three replicates in each group.Wedetected the changes in malondialdehyde（MDA） content on the1st，3rd and 5thdays，as wellas sperm motilityvitality，plasma membrane integrityand acrosome ntegrityfromOto5days.Theresults showed that with theextension of storage time，sperm motility，activity，and motility gradually decreased，and MDA content gradually increased. The motility and activity of the 10μmol⋅L-1 group from the 2nd to the 5th day were significantly higher than those of the other groups （ ⋅P<0.05 ）；the VSL of the control group from the 3rd to the 5th day，and the VCLand VAP from the2nd to the 5thday were significantly lower than those of the 10μmol⋅L-1 （204号 group （ .P<0. 05 ）； the plasma membrane and acrosome integrity rates of the 10μmol⋅L-1 group throughout the process were significantly higher than those of the control group （ P<0.05 ）；the MDA content of the 10Ωμmol⋅L-1 group was also significantly lower than that of the other groups （ P<0.05 ）.The experimental results indicated that adding a certain proportion of IDE to the diluent could improve the qualityof Hu sheep sperm，aleviate the damage to the sperm membrane，and achieve the effect of improving the quality of Hu sheep semen. When 10Ωμmol⋅L-1 IDE was added to the cryopreservation diluent，the preservation effect of Hu sheep semen was the best.
