ABSTRACT:To explore the mechanism of nobiletin(NOB)in the treatment of postpartum depression using network pharmacology,molecular docking and animal studies.The modelof postpartum depresion was established and the pharmacological efects of NOB were evaluated by behavior analysis.The targets ofnobiletin,postpartum depression,estrogen withdrawal,and inflammation were obtained from databases such as TCMSP.Protein-protein interaction network was generated using STRING database,and Cytoscape software topology analysis was used to examine the network. Metascapedatabase wasapplied for GO and KEGG enrichment analysis,and AutoDock Vina was used to complete the molecular docking.Results showed that medium and high doses of NOBsignificantly improved postpartum depression-like behaviors inmice.A total of 2O6 intersection genes were obtained by network pharmacological analysis. Core signaling pathways included PI3K-AKT,MAPKand estrogen signaling pathway,etc.Moreover,eytargets had good binding activityto NOB.By acting on AKT1,ESR1,BCL2 and others,NOB regulated the PI3K-AKT,MAPK and estrogen signaling pathways to aleviate postpartum depression. This study suggests that NOB may be a potential treatment for postpartum depression.
